Iovance Biotherapeutics Inc. (Nasdaq: IOVA) has reported updated Phase 2 results for lifileucel in patients with metastatic non-small-cell lung cancer whose disease progressed following chemotherapy and immune-checkpoint treatment.
As of the October 14, 2026 data cutoff, 17 of 69 patients responded to treatment, producing an objective response rate of 24.6%. Fifteen responses were confirmed and two were unconfirmed partial responses awaiting additional assessment.
The results came from the non-squamous lung-cancer groups in the open-label IOV-LUN-202 study. Patients receive a lymphodepleting regimen followed by an infusion of their own tumor-infiltrating lymphocytes, or TILs, and subsequent interleukin-2 treatment.
A retrospective biomarker analysis found substantial differences between patient subgroups. The response rate was 43.3% among 30 patients with KRAS wild-type tumors, compared with 11.4% among 35 patients with KRAS-mutant disease.
Iovance further identified a 22-patient subgroup combining KRAS wild-type status, HER2 wild-type status and a history of smoking. Thirteen of those patients responded, including two complete responses, resulting in a 59.1% response rate.
“The expanded dataset reinforces the activity of lifileucel in previously treated disease,” interim President and CEO Frederick Vogt said. Management believes the subgroup findings could support a more selective approach to identifying patients for TIL therapy.
The 59.1% result should be interpreted cautiously. The criteria were applied retrospectively rather than established before patients entered the trial, and the subgroup included only 22 people. The study also lacks a randomized control group, while two of the 17 responses reported across the broader population remain unconfirmed.
Iovance plans to test the combined biomarker strategy prospectively through a new cohort called POSITILITY. Enrollment is expected to begin in November at the first of more than 70 existing IOV-LUN-202 sites. The company intends for the study to support a potential registration pathway, subject to FDA feedback.
The broader trial is expected to include approximately 80 non-squamous lung-cancer patients in its final dataset. Iovance is planning a supplemental Biologics License Application for lifileucel in this indication during 2027.
Safety remains a material consideration. The FDA placed IOV-LUN-202 under a clinical hold in December 2023 following a fatal serious adverse event potentially related to the lymphodepletion regimen. The agency lifted the hold in March 2024 after Iovance reduced the lymphodepletion dose. The latest announcement said the revised regimen improved the overall safety profile but did not provide detailed updated safety figures.
Lifileucel is already marketed as Amtagvi for certain previously treated melanoma patients but remains investigational in lung cancer.
Iovance reported second-quarter product revenue of $99.3 million, a net loss of $47.3 million and approximately $303.7 million in cash, investments and restricted cash as of June 30, 2026. The company estimates that its capital can support operations into the second half of 2028.

Sources used include Iovance’s October 29, 2026 clinical-data announcement, ClinicalTrials.gov, FDA materials and the company’s second-quarter results and Form 10-Q. Information concerning lifileucel’s clinical performance, biomarker strategy and planned regulatory path originated primarily from Iovance and has not been independently validated by Biotech Stocks Daily.
Lifileucel remains investigational for non-small-cell lung cancer. The reported findings come from an open-label, non-randomized study. The 59.1% response rate was observed in a retrospectively selected subgroup of 22 patients and may not be reproduced prospectively. Two responses included in the broader objective response rate remain unconfirmed.
The IOV-LUN-202 trial was previously placed under a clinical hold following a fatal serious adverse event potentially related to the lymphodepletion regimen. Although the FDA allowed enrollment to resume using a reduced regimen, additional serious adverse events, clinical delays or regulatory requirements remain possible.
Financial information reflects Iovance’s position as of June 30, 2026. The company reported an accumulated deficit of approximately $2.9 billion and expects additional operating losses. Its projected cash runway into the second half of 2028 is a forward-looking estimate that depends on commercial performance, clinical spending and other assumptions.
